以苋菜全基因组数据为基础,共鉴定出21个Hsf基因家族成员,分布在14条染色体上,其氨基酸残基数256~535个,分子量为29.71~59.73 kD,等电点为4.69~9.22,19个酸性蛋白,2个碱性蛋白,20个亲水性不稳定蛋白,1个亲水性稳定蛋白,定位于细胞核上。AtrHsf基因家族成员结构较为保守,启动子区具有大量环境及激素响应元件,成员间具有明显的共线性。高温转录组数据表明,大部分AtrHsf基因受高温诱导,表现出不同的表达模式,qRT-PCR结果与转录组结果基本一致。该研究为苋菜高温胁迫下Hsf基因的调控机制分析奠定了基础,为耐高温种质资源创制及新品种选育提供了重要基因资源和理论基础。
Abstract:Based on the whole-genome data of amaranth, this study identified 21 members of the Hsf gene family, which are distributed on 14 chromosomes.The number of amino acid residues ranges from 256 to 535,the molecular weight ranges from 29.71 to 59.73 kD,and the pI ranges from 4.69 to 9.22.There are 19 acidic proteins, 2 basic proteins, 20 hydrophilic and unstable proteins, and 1 hydrophilic and stable protein, all of which are located in the nucleus.The members of the AtrHsf gene family have a relatively conserved structure.The promoter regions have a large number of environmental and hormone response elements, and there is obvious collinearity among the members.High-temperature transcriptome data show that most of the AtrHsf genes are induced by high temperature and exhibit different expression patterns.The results of qRT-PCR are basically consistent with those of the transcriptome.This study lays a foundation for the analysis of the regulatory mechanism of Hsf genes in amaranth under high-temperature stress, and provides important gene resources and a theoretical basis for the creation of heat-tolerant germplasm resources and the breeding of new varieties.
