为提升雪茄茄衣烟叶的发酵质量,引入外源微生物腐生葡萄球菌(Staphylococcus saprophyticus),并与自然发酵组进行对照分析。通过对不同发酵处理的烟叶样本进行挥发性香气成分分析和宏基因组分析,研究腐生葡萄球菌在雪茄烟叶发酵中的应用及其对微生物群落、香气成分和代谢通路的影响。结果表明,腐生葡萄球菌的加入明显提高了挥发性香气成分的含量。宏基因组分析显示,腐生葡萄球菌引入对微生物群落产生影响,但芽孢杆菌属(Bacillus)仍为主导。KEGG分析表明,试验组在碳水化合物代谢、氨基酸代谢等通路中活性明显增强。EGGNOG分析进一步揭示了试验组中与细胞外结构、信号转导机制、细胞运动相关基因表达上调。CAZy分析表明,试验组糖基转移酶(GT)、碳水化合物结合模块(CBM)和碳水化合物酯酶(CE)相关基因的相对丰度增加,促进了碳水化合物代谢。LEfSe分析发现了不同处理组的差异微生物;Spearman相关性分析显示,葡萄球菌属与芽孢杆菌属呈负相关,与多种关键香气成分呈正相关。

Abstract:

To enhance the fermentation quality of cigar wrapper leaves, Staphylococcus saprophyticus was introduced as an exogenous microorganism and compared to a naturally fermented control group. Through the analysis of volatile aroma components and metagenomic analysis of tobacco leaf samples with different fermentation treatments, the application of Staphylococcus saprophyticus in the fermentation of cigar tobacco leaves and its effects on microbial communities, aroma components and metabolic pathways were studied. The results demonstrated a significant increase in the content of volatile aroma compounds following the addition of Staphylococcus saprophyticus. Metagenomic analysis revealed that while the introduction of Staphylococcus saprophyticus influenced the microbial community, Bacillus remained the dominant genus. KEGG pathway analysis indicated a marked enhancement of activities in carbohydrate and amino acid metabolism pathways in the experimental group. EGGNOG analysis further highlighted upregulation of genes related to extracellular structures, signal transduction, and cell vitality. CAZy analysis showed an increase in the related genes' relative abundance of glycoside transferases(GT), carbohydrate-binding modules(CBM), and carbohydrate esterases(CE), promoting carbohydrate metabolism. LEfSe analysis identified significantly different microbial populations across treatments, while Spearman correlation analysis revealed a negative correlation between Staphylococcus and Bacillus, both of which were positively correlated with various volatile aroma compounds.
