摘要:

[目的]获得高产脂肪酶蜡样芽孢杆菌(Bacillus cereus),优化产酶菌株培养条件。[方法]选取68株蜡样芽孢杆菌为试验菌株,以酶活为指标,利用Tween-80平板定性筛选产酶菌株,对高产脂肪酶菌株通过单因素试验和Plackett-Burman试验筛选影响粗酶液酶活的显著因素,最陡爬坡试验和响应面法探究最佳发酵工艺。[结果]胰蛋白胨、磷酸氢二钾、七水合硫酸镁是影响野生型蜡样芽孢杆菌产脂肪酶的主要因素,其最适添加量分别为11.13、1.59、1.51 g/L,优化后酶活可达85 U/mL。[结论]此方法适用于探究影响N447-1产酶的主要因素和最适添加量,优化后产酶能力显著提升。

Abstract:

[Objective]To optimize the culture conditions for enzyme-producing strains to enhance lipase production in Bacillus cereus.[Method]68 Bacillus cereus strains were utilized as experimental subjects.And Tween-80 plates were employed for qualitative screening of enzyme-producing strains.Significant factors affecting crude enzyme activity were identified through single-factor experiments and Plackett-Burman design, followed by the optimization of fermentation parameters using the steepest ascent methodology and response surface analysis.[Result]Tryptone, dipotassium phosphate, and magnesium sulfate heptahydrate were identified as primary factors influencing lipase production in wild-type Bacillus cereus.Optimal concentrations were determined as 11.13,1.59,and 1.51 g/L,respectively, achieving enzyme activity of 85 U/mL after optimization.[Conclusion]This systematic approach effectively identifies critical factors and optimizes supplementation levels for strain N447-1,demonstrating a significant enhancement in enzymatic productivity.
