石渠棘球绦虫Es95基因克隆及序列分析Cloning and Sequence Analysis of Es95 Gene from Echinococcus shiquicus

摘要(Abstract):
[目的]首次克隆获得石渠棘球绦虫Es95基因,并进行序列分析,为研究抗包虫病候选疫苗提供理论指导。[方法]从高原鼠兔肺脏包囊上获得石渠棘球绦虫原头蚴,提取全基因组,并根据NCBI上6种棘球属绦虫95基因序列设计引物,以基因组DNA为模板扩增到Es95基因,并克隆到T载体上,测序后进行序列分析。[结果]以石渠棘球绦虫全基因组DNA为模板扩增出Es95基因长为1 330 bp,序列分析含有3个外显子和2个内含子,有1个糖基化位点,N端有一信号肽序列(16个氨基酸),C端有跨膜区(23个氨基酸),亲水性低,B细胞抗原表位预测有ES95潜在抗原表位6个。[结论]ES95是一个分泌性糖蛋白,在包虫病的预防上具有重要意义。
