有机相中Novozyme435催化的丁基苯酞的有效转酯化Research on Effective Transesterification of 3-N-butylphthalide Catalyzed by Novozyme 435 in Organic Phase

摘要(Abstract):
[目的]研究在非水相介质中脂肪酶催化丁基苯酞的转酯化可行性。[方法]以脂肪酶Novozyme435、PPL、PS、PS-C和AY为试材,通过酶法反应试验和对反应后丁基苯酞的转化量进行气相色谱分析,研究了某些关键因素如四氢呋喃(THF)、有机溶剂、含水量以及丁基苯酞与乙酸乙烯酯的摩尔比等对转酯化反应的影响。[结果]THF有助于改善水在疏水性介质中的均匀性分布,增加水在疏水性溶剂中的溶解度而降低反应介质的水活度,进而大大提高丁基苯酞在微水相有机溶剂中的转酯化得率。在该反应中,由65%(mol/mol)的THF和35%(mol/mol)的正己烷组成的二元混合溶剂为适宜的反应介质,最适含水量为0.4%(V/V),在该条件下,Novozyme435催化的转酯化可使丁基苯酞的转化率达到49.4%。[结论]脂肪酶Novozyme435对催化丁基苯酞的转酯化具有较高的催化活力。
