噬菌体P1侵染大肠杆菌Nissle 1917(EcN)机理研究Study on the Mechanism of Phage P1 Infecting Escherichia coli Nissle 1917 (EcN)

摘要(Abstract):
[目的]找出噬菌体侵染大肠杆菌的位点。[方法]选择血清型为O6∶K5∶H1的大肠杆菌Nissle 1917(Ec N)作为试验菌株。通过λ-RED系统,突变其K抗原和O抗原合成基因,通过检测P1转导至这一系列突变株的转导效率来确定P1的侵染位点。[结果]双突变株Ec NΔkfi AΔwaa G::Cm具有最高的转导效率。kfi A基因突变导致了K抗原缺失,waa G基因突变导致了LPS核心寡糖中第2位庚糖(HepⅡ)的暴露,使得P1更容易接触到HepⅡ,从而侵染细菌。[结论]研究认为HepⅡ很可能就是噬菌体P1的识别位点。
