苹果多酚对高糖高脂膳食加STZ诱导的糖尿病小鼠血脂·血糖水平和PPARγ基因表达的影响Effects of Apple Polyphenol on Blood Lipid,Blood Glucose Level and PPARγ Gene Expression of Diabetic Mice

摘要(Abstract):
[目的]研究苹果多酚对糖尿病小鼠血脂、血糖水平和肝、肾、脂肪组织PPARγmRNA表达的影响。[方法]雄性小鼠分为对照组(n=12)、模型组(n=12)和苹果多酚干预组(n=36)。通过给予高脂高糖饲料喂养,腹腔注射链脲佐菌素,建立糖尿病小鼠模型。应用苹果多酚对糖尿病小鼠干预治疗60 d。在试验结束时,集中处死鼠,搜集血标本和肝、肾、脂肪组织标本。检测各组大鼠的血MDA、FBG、FINS、TG、TC等浓度。采取RT-PCR方法,检测各组小鼠肝、肾、脂肪组织PPARγmRNA的表达。[结果]与Con组相比,模型组小鼠血清MDA、血脂、血糖、胰岛素水平显著升高(P<0.01),与模型组相比,苹果多酚能显著降低小鼠的血清MDA、血脂、血糖、胰岛素水平(P<0.05)。与Con组相比,模型组小鼠肝、肾、脂肪组织PPARγmRNA表达水平显著下降,苹果多酚组小鼠肝、肾、脂肪组织PPARγmRNA的表达水平较模型组均有明显增加(P<0.05)。[结论]苹果多酚能够降低糖尿病小鼠血清MDA、血脂、血糖水平,减轻胰岛素抵抗,提高PPARγmRNA的表达。苹果多酚可能发挥着类似PPARγ激动剂的效用,通过提高PPARγmRNA的表达来调节糖尿病小鼠的血糖、血脂代谢。
