用ISSR标记分析甜荞栽培品种的遗传多样性Analysis on Genetic Diversity in Common Buckwheat(Fagopyrum esculentum Moench) Based on ISSR Markers

摘要(Abstract):
[目的]研究用ISSR标记分析甜荞栽培品种的遗传多样性。[方法]采用ISSR分子标记对来自11个省的90份甜荞种质的遗传多样性进行了分析。[结果]19条ISSR引物共扩增出508条条带,平均26.7条;其中多态性条带462条,平均24.3条,多态性带的百分率为90.1%。Nei’s遗传相似系数在0.67~0.95。UPGMA聚类分析显示,聚类结果与地理来源有较强的一致性,来自辽宁、内蒙古、陕西、四川、甘肃等省的甜荞都是按来源各自聚为一类,但是发现了几个特殊的类型,即来自河北的甜荞E、安徽的82F以及辽宁的辽荞37号均单独聚为一类。不同地区材料之间的多态性信息指数(PIC)差异较大,其中,辽宁的最高,为0.842,内蒙古的最低,为0.633。[结论]采用ISSR分析甜荞遗传多样性,甜荞表现出丰富的多态性,表明可以用ISSR标记对甜荞进行分子水平的鉴定和遗传多样性分析。
