小麦赤霉菌FGSG_03700基因敲除及功能研究Research on Knock-out and Function of FGSG_03700 Gene in Fusarium graminearum

摘要(Abstract):
[目的]敲除小麦赤霉菌(Fusarium graminearum)FGSG_03700基因,确定其缺失突变体表型,从而分析该基因的生物学功能。[方法]应用Split Marker基因敲除技术,构建含有潮霉素抗性基因hph的基因敲除盒,通过PEG介导,进行原生质体转化,在含有潮霉素的培养基上筛选转化子。[结果]通过PCR正负筛查,得到3个确定的突变体,分别命名为ΔFGSG_03700 1-2、ΔFGSG_03700 2-2、ΔFGSG_03700 2-3。通过表型验证及孢子对番茄侵染试验发现,敲除突变体的菌落形态及生长速度没有明显变化,致病力没有减弱,但突变体的产孢量低于野生型PH-1。[结论]FGSG_03700基因可能与小麦赤霉菌分生孢子的生长能力有关。
